The key to cancer therapy utilizing microbes is the ability to track the
spread and replication of the bacteria or viruses within the body over
time. A number of strategies employing IVIS^® optical imaging for
tracking reporter constructs in different models, from /in vitro/
through to clinical samples have been developed. For bacterial work,
novel /lux/ reporter constructs for luminescence imaging of different
genera /in vitro/ and /in vivo/ in mice have been generated. This has
permitted 2D and 3D whole body imaging for a wide range of both invasive
and non-invasive species in different anatomical settings– in tumors,
various deep organs, and even within the gastrointestinal tract of live
animals.
For eukaryotic work (tumors or viruses), luciferase tagged constructs
permit similar imaging, the ability to monitor different luminescent
reporters simultaneously (e.g. lux + Fluc within the same tumor),
providing highly detailed monitoring of infections and host responses
has been demonstrated.
Additionally, techniques for luminescence imaging of microbes and gene
delivery in patient tissue /ex vivo/ to provide clinically relevant data
have been developed.
